Whether there is an opportunity of such optimization? If isn't present, why? If yes, there is at somebody an source code and he can share it? As I have understood under the description of algorithm, the algorithm is based on work with DWORDs. It is a correct conclusion?

It is based on DWORDS, however, the expanded register set of x64 makes it much simpler to calculate MD5 sums.
